Nanobiotechnology represents a powerful convergence of nanotechnology and biotechnology, showing innovative approach in diagnosis, treatment, and drug delivery. Among all transformative applications, smart drug delivery systems that utilize nanomaterials for precise and controlled therapeutic action stands most useful. These nanoscale particles, that minimize systemic adverse effects while enhancing pharmaceutical efficacy, include liposomes, dendrimers, polymeric nanoparticles, metallic nanoparticles, and carbon-based nanostructures. These allow guided drug transport to particular tissues or cells. The design of such nanomaterials is characterized by parameters such as size of nanopartices, surface charge, and functionalization, this effects their interaction with biological environments and their ability to overcome physiological barriers.

Site-specific drug release and real-time therapeutic control are made possible by smart nanocarriers’ ability to react to both external and internal stimuli, including as temperature, light, magnetic field, pH, redox potential, and enzyme activity. Compared to traditional delivery methods, which frequently have poor bioavailability and off-target effects, this responsive behavior represents a significant development. Additionally, nanobiotechnology facilitates the co-delivery of multiple drugs or imaging agents, integrating diagnosis and therapy into a single platform.

With these developments, clinical application still faces obstacles like large-scale synthesis, long-term toxicity, biodegradability, and regulatory uniformity. Developing sustainable, multipurpose nanocarriers and investigating customized nanomedicine for patient-specific therapies are the main areas of current study.

This chapter discusses the recent progress, mechanisms, and future prospects of nanomaterials in smart drug delivery, highlighting how nanobiotechnology is redefining the next generation of advanced therapeutics.
